The folin phenol reagent method consists of two steps: the first step is protein interact with copper to form protein-copper complex in an alkaline solution. The second step is the complex reduce the Folin reagent to produce dark blue. The amount of color produced is proportional to protein concentration. The quantitative rang of protein is 5~100μg/ml. Folin reagent chromogenic reaction is caused by tyrosine, tryptophan and cysteine. Therefore, phenols, citric acid and sulfhydryl compounds in the sample have interfere. Besides, different proteins have different color intensity due to different contents of tyrosine and tryptophan. Lowry法包括兩步反應(yīng):第一步是在堿性條件下,蛋白質(zhì)與銅作用生成蛋白質(zhì)-銅絡(luò)合物;第二步是此絡(luò)合物將Folin試劑還原,產(chǎn)生深藍(lán)色,顏色深淺與蛋白質(zhì)含量成正比。定量范圍為5~100μg/ml蛋白質(zhì)。Folin試劑顯色反應(yīng)由酪氨酸、色氨酸和半胱氨酸引起,因此樣品中若含有酚類(lèi)、檸檬酸和巰基化合物均有干擾作用。此外,不同蛋白質(zhì)因酪氨酸、色氨酸含量不同而使顯色強(qiáng)度稍有不同。 |
